Everything We Know About Managing Pain With Cannabis

Though studies have found that CBD is invaluable in helping manage certain seizure disorders and that THC is a near-miracle treatment for those suffering from nausea and loss of appetite, there is one issue that cannabis users always come back to: pain management.

Chronic pain is one of the worst diagnoses imaginable. Often, the source of chronic pain is difficult for medical professionals to identify, which makes it almost impossible to treat effectively. Current pain management solutions include opioids and NSAIDs, both of which can have devastating impacts on health and well-being in the short and long term. Thus, it makes sense that natural, safe cannabis is rapidly becoming a popular pain management solution for those suffering from chronic pain.

Yet, the jury is still out with regards to whether cannabis does actually positively affect the experience of pain. Here’s a rundown of current research regarding pain relief and cannabis, so you can make an informed decision for your health.

Pain Reception and the Endocannabinoid System

You might believe that pain reception is solely the realm of the nervous system. Nerves throughout your body help you sense and experience the world around you, so why wouldn’t nerves be responsible for detecting and communicating pain? It seems that it is slightly more complicated than that.

Closely associated with the nervous system — and several other systems within the body — is the endocannabinoid system, or ECS. The ECS has many responsibilities essential for maintaining life, chief among them being the management of homeostasis, or internal balance despite external change. To accomplish this, the ECS can shift core temperature, moderate metabolism, elicit stress responses and, most germanely, signal pain.

Since discovering that the endocannabinoid system is affiliated with the experience of pain, many studies have sought to better understand its role. Though knowledge surrounding this issue continues to evolve, at present researchers believe that the ECS has control over blocking or permitting the sensation of pain. It does this by creating a compound called palmitoylethanolamide, or PEA, which binds to receptors in the nervous system to send messages about pain to the brain. PEA has analgesic properties, meaning it essentially blocks the experience of nociceptive pain, or pain that develops as the result of physical stimuli, like a hot pan or a strained muscle.

There is another type of pain, called neurological pain, that results from a malfunctioning nervous system possibly caused by damaged nerves. Though less is understood about the ECS’s role in moderating neurological pain, there is some evidence that endocannabinoids can impact this pain experience as well.

Analgesia Through THC and CBD

Through history, marijuana has been used as a tool for managing pain, and many sufferers of chronic pain vociferously argue that weed is one of the only truly effective treatments for their condition. As a result, research has sought to better understand what compounds within cannabis function as analgesics and how.

Unfortunately, science has not yet revealed all of these secrets. Currently, the two most-studied compounds unique to cannabis are the cannabinoids delta-8 tetrahydrocannabinol / delta-8 THC, and cannabidiol, or CBD.

THC is the dominant compound responsible for getting marijuana users high. It binds directly to the endocannabinoid system’s CB1 receptors, which are located primarily in the brain and digestive system. In contrast, researchers aren’t certain how CBD interacts with the human body; it seems to encourage either the ECS or the nervous system to release endocannabinoids or neurotransmitters.

Both THC and CBD can be valuable in mitigating the experience of pain. THC has an almost identical structure to the neurotransmitter anandamide, also known as the bliss molecule. Thus, those suffering from chronic pain can use THC as a distraction for their brains, a way to feel good. Further, there is evidence that the endocannabinoids and/or neurotransmitters released by CBD help eliminate inflammation and block pain temporarily, which can be an immense relief.

Conclusion

Science will continue to reveal more about how the ECS interacts with pain expression and how compounds with cannabis can be applied to make chronic pain more manageable. Pfizer × BioNTech near historic vaccine

Pfizer and BioNTech announced on Monday that their COVID-19 vaccine is more than 90 percent effective. If approved, it could potentially be available to the public by early December, according to Dr. Anthony Fauci.

The results came out of Pfizer’s Phase 3 trial, which involved 40,000 individuals. Of those participating, 94 contracted COVID-19. These results, like much of 2020, are historic. Vaccines have never been developed on such a fast-moving timeline. The last vaccine that was developed in such considerable haste was for mumps, and it took four years.

Pfizer says that they plan to ask the Federal Drug Administration for emergency use by the end of the month. The vaccine will require two doses administered three weeks apart. The company hopes to have enough doses for 25 million people by the end of the year, and 650 million people in 2021.

In the case that the vaccine supply is limited, the C.D.C. will first vaccinate healthcare personnel, essential workers, people who are at high risk for severe COVID-19 illness due to underlying medical conditions, as well as those 65 years and older.

An expedited timeline does not mean drug companies are cutting corners. Fauci, like many of his colleagues in Washington, assures that manufacturers will stick to a process of vaccine development that ensures the safety of patients. The FDA will still make the final call.

“Today is a great day for science and humanity,” said Dr. Albert Bourla, Pfizer Chairman and CEO, in Pfizer’s press release. “The first set of results from our Phase 3 COVID-19 vaccine trial provides the initial evidence of our vaccine’s ability to prevent COVID-19.”

However, a few questions remain unanswered.

“Historically, important scientific announcements about vaccines are made through peer-reviewed medical research papers that have undergone extensive scrutiny about study design, results and assumptions,” writes Arthur Allen in the Opinion section of the NYT. “Not through company press releases.”

According to Allen, it is unclear from the press release how long Pfizer’s vaccine will keep patients protected, if it is safe for high-risk populations like the elderly, or if rare side effects can arise in patients who are vaccinated. He notes that the Novavax and Sanofi Pasteur vaccines may be safer for older patients.

Novavax and Sanofi Pasteur are subunit vaccines, like the hepatitis B vaccine. They deliver only the essential antigens of the virus to the immune system, so it learns how to attack it. Because it is only a part, or a subunit, of the virus, fewer side effects are likely.

Pfizer’s is a nucleic acid vaccine that uses RNA. According to the Washington Post, “this type of vaccine contains a strip of genetic material within a fat bubble” that enters the cell. Once inside, “the RNA generates a protein found on the surface of the virus.” It can then familiarize itself with the virus and learn how to fight it.

Moderna’s vaccine is also an RNA vaccine in Phase 3 trials. Pfizer’s success bodes very well for Moderna, according to a statement Fauci made to CNN.

Furthermore, because the vaccine must be stored in extremely low temperatures—on dry ice at negative 100 degrees Fahrenheit according to the Washington Post—its roll-out becomes complicated. If left out in the sun, or just at room temperature, or even at just below freezing, the mRNA self-destructs and the vaccine becomes useless.

Shortly after Pfizer’s announcement, President-elect Joe Biden addressed the nation, warned of the “dark winter” ahead, and urged Americans, regardless of party affiliation, to wear a mask until the vaccine is available.

The head of the C.D.C. warned this fall, that “for the foreseeable future, a mask remains the most potent weapon against the virus,” he said from the podium. “Today’s news does not change that urgent reality.”

Gilda’s Club x The Yellow Cab Project

Nov. 12, the Gilda’s Club NYC will host their annual gala, honoring Giovanni Caforio (M.D. Bristol Myers Squibb) and Emma Stone (Actress and Gilda’s Club NYC Ambassador), virtually.

As part of this year’s Gilda’s Club NYC fundraising, Remembering Marco will participate in the virtual auction launched on Nov. 5 with The Yellow Cab Project. The Yellow Cab Project is part of Remembering Marco, a charity initiative started by Annalisa Menin in 2013 to raise funds in memory of her husband, Marco Omiccioli.

Annalisa Menin, an Italian entrepreneur and writer who was originally from Venice, Italy, built her name with her first publishing project “My Last Year in New York,” a blog that then became a book.

It was dedicated to her husband, Marco Omiccioli, who died prematurely at the age of 33 from cancer in Nov. of 2013.

The Yellow Cab Project started when Annalisa stopped in front of the Kendra Scott window in SoHo, located at the intersection of Spring and Greene Streets. Along with jewelry and beautiful handbags, there were a few die-cast model yellow cabs being used as props, a tribute to the city of New York.

Annalisa went inside to ask what would happen to all those models once the window was emptied. Once she discovered that they would be thrown away, she thought, “Oh, no! I want them!”

Two weeks later, she went home in a real life version of those models, driven by a lovely gentleman with a white, cloud-shaped beard. She carried eight boxes containing as many cabs.

Annalisa created The Yellow Cab Project in 2020. It involved five international artists, designers and creatives, giving them the opportunity to “contaminate” the die-cast model cabs in their own way. The five artists were Warø, Jon Koon, Iena Cruz, Marco Gallotta and Clint Henderson.

To remember Marco in this very eventful year, Annalisa chose to support Gilda’s Club NYC. She strongly believes in their mission to “support, educate, and empower cancer patients and their families.”

This year, Gilda’s Club is turning 25 years old, and they will celebrate this important milestone with a virtual gala Nov. 12. It is the perfect occasion for the die-cast models, now precious objects of beauty and art, to be donated and auctioned, raising funds for Gilda’s Club NYC and its community from Nov. 5 to Nov. 16.

“We are so excited to have these one-of-a-kind yellow taxis included in our 25th Anniversary milestone event to benefit our cancer patients and families and are grateful for this generous collaboration with Annalisa and the remembering Marco Omiccioli Charity Initiative,” said Lily Safani, CEO of Gilda’s Club.

In these unprecedented times of the COVID-19 pandemic, Gilda’s Club NYC, the cancer support organization founded in memory of Saturday Night Live comedienne, Gilda Radner, transitioned their free cancer support program to a virtual program. They will continue to provide essential support services to cancer patients who are among some of the most vulnerable in this pandemic.

Members have expressed sincere gratitude for their quick transition to virtual programming. Due to the increased demand for this program and an affected donor base, they need additional funds to serve their current and growing community.

Annalisa personally experienced how important it is for grieving individuals to have a support system, especially in a city like New York, where many are far away from loved ones. She joined one of the bereavement group at Gilda’s Club NYC in 2013 after her husband Marco passed away, and it helped her tremendously.

The Bucket List

Positioned on the Adriatic Sea, Croatia is a country at the intersection of Central and Southeast Europe. Therapies Developed to Reduce Lung Fibrosis

A new treatment option for lung fibrosis is being developed by Purdue University scientists. Lung fibrosis has been a concern for COVID-19 patients.

People with idiopathic pulmonary fibrosis (IPF) have a life expectancy of fewer than five years. Fibrotic diseases cause organ failure that leads to about 45% of all deaths in the United States. Existing therapies do little to slow progression.

Now, Philip S. Low, Purdue’s Ralph C. Corley Distinguished Professor of Chemistry and Presidential Scholar for Drug Discovery, has led a team to develop two targeted therapies for people with IPF. The two different therapeutic approaches are published in Science Translational Medicine and EMBO Molecular Medicine.

“This is a horrible disease that claimed the lives of my next-door neighbor and a good friend’s wife,” Low said. “We developed two targeted therapies that allow us to use powerful drugs with high toxicities because we specifically deliver them to diseased cells without harming healthy ones.”

The first of the Purdue team’s novel targeted molecules is designed to slow fibrosis and extend life. The second IPF therapy suppresses fibrosis-inducing cytokine production.

The two therapies will be moving into human clinical trials within the next several months. The developments come as a number of people with COVID-19 or who have recovered from COVID-19 experience lung fibrosis or other related conditions.

His mother was a singer, almost signed by Quincy Jones, but she later became a teacher to have more time to focus on raising her son, but still kept the music alive within him. He would listen to his mom’s favorites like Mary J. Blige, Michael Jackson, Jodeci, and Usher, while tempering them with New York City street rap. He studied battle rap, fascinated by its competitive nature, and likened it to the sports he played in school. When delivering water to a recording studio one day, it ended up being to Diddy at Daddy’s House. Ryan met Diddy’s son Christian (bka King Combs) and told him that he was a rapper and they became fast friends. Ryan ultimately started building his resume, networking with top R&B and Hip-Hop artists and executives. He was then fortuitously introduced to industry titan Wayne Barrow, who introduced him to his business partner, fellow industry veteran Mark Pitts, which then led to his signing to StormTroopers/RCA Records.

With his own music, Ryan’s focus gradually shifted more from rapping to singing, no doubt influenced by his ability to write for other artists, while still maintaining the spirit of Hip-Hop. He played his track “Real One” for Chris Brown and Jeremih and they encouraged him to release it. The track became the catalyst that sparked the next steps in his career. He’s got inevitable soul, mixed with ambition and an edge that is a clear testament to his upbringing. His recently released “Back Up” has hints of Caribbean flavor, a nod to Ryan’s Brooklyn upbringing, but also his cultural background (he’s Jamaican, African American, and Puerto Rican). The video comes complete with coordinated dances, reflecting a true dancehall vibe. Ryan’s debut EP Games We Play, out now via StormTroopers/RCA Records,  is an amalgam of every element of R&B and Soul music from past to present. The meaning is represented by the title track, as the game of love comes with many pieces.

Bugatti Baby II

The Bugatti Owners’ Club and The Little Car Company announced a new partnership Wednesday following the release of The Little Car Company’s Bugatti Baby II.

With this partnership, the Bugatti Owners’ Club gives its stamp of approval to the Bugatti Baby II, giving it certified status and hopefully building the foundation for a long and successful relationship.

Based at the Prescott Hill Climb in the UK, the Bugatti Owners’ Club was founded in 1929 and has more than 2,000 members.  It is the oldest club for the marque in the world.

This partnership will allow members of the two organizations to connect and share their passions for cars, design and engineering. It will also offer the chance for members to get together at events at the Bugatti Owners’ Club clubhouse.

The Bugatti Baby II is a remastering of the original Bugatti Baby. It preserves the history of Bugatti while also inspiring a new generation of Bugatti fans and enthusiasts.

Gemma Price, the general manager of the Bugatti Owners’ Club said she is delighted to work with The Little Car Company.

“We are looking forward to working with The Little Car Company closely and developing a strong partnership centered around the excellence of Ettore Bugatti’s design and engineering prowess that has proven to be visionary and timeless throughout the decades,” Price said. “This has been exceptionally captured and reimagined with the Bugatti Baby II and we look forward to celebrating this common affection through the development of the association of our two organizations.”

The first Bugatti Baby was created in 1926. Ettore Bugatti and his son decided to build a half-scale version of he Type 35 for Ettore’s youngest son’s fourth birthday. It was expected to be a one-off, but it received positive feedback, so the car went into production. 500 units were made between 1927 and 1936.

The Bugatti Baby II is based on a digital 3D scan of a Lyon GP car, so the handling is authentic. The handling can be adjusted because of adjustable dampers, which you can see an example of by clicking right here.

It also has replicas of the original Type 35’s instruments but with modern practicality. The fuel pressure gauge is now a battery gauge. Also, the oil gauge has been transitioned to a power gauge, which is a nod to the Bugatti Veyron.

It is also 75% scale of the Type 35 and can go from zero to 60 kph in six seconds, depending upon the weight of the driver. It tops out at 70 kph, and only 500 will be made.

As the 1982 recipient of the Ford C. Frick Award, which enshrined him in the broadcasters’ wing of the Baseball Hall of Fame, Scully has covered many of baseball’s most thrilling moments, including Sandy Koufax’s four no-hitters, Hank Aaron’s 715th home run and Kirk Gibson’s dramatic home run in Game One of the 1988 World Series. He has voiced over the narration for seven previous World Series documentaries including in 1959 (Dodgers), 1963 (Dodgers), 1965 (Dodgers), 1979 (Pirates), 1980 (Phillies), 1981 (Dodgers) and 1986 (Mets).

It was 32 years since the Dodgers won a World Series and the seventh in the history of the organization, bringing great joy to their fans and the city of Los Angeles in particular, said Scully. This is their winning story against the Tampa Bay Rays, and it’s an honor for me to recount just how they did it.

With MLB playing in a postseason bubble, media presence and access was limited.
